Preservation of Viable Leucocytes in Glycerol at −80° C.

Abstract

SINCE the demonstration by Polge et al.1 of the protective effect of glycerol on fowl spermatozoa stored at low temperatures, a number of mammalian tissues, including erythrocytes and bone marrow cells of man, have been successfully preserved in this manner2–4. With the recent interest in the culture of leucocytes from peripheral blood in determining human karyotypes, it would be desirable to be able to preserve leucocytes at low temperatures for later study or to permit shipment over long distances.
