Labelling Rates and Detection of Intermediates in Mitochondrial Phosphorylations and other Sequential Reactions

Abstract

USE of isotopes frequently provides a sensitive tool for detection of small concentrations of intermediates in metabolic sequences. Valuable information about the possible participation of an intermediate may often be obtained by measurement of rates of appearance of isotopic tracer in a possible intermediate and in a subsequent metabolic product. Such approaches have recently been applied to the assessment of possible phosphorylated intermediates in the formation of ATP by mitochondria, but important differences in the interpretation of the results have arisen1–3.
