Powerful New Aggregator of Blood Platelets—2-Chloroadenosine-5′-diphosphate

Abstract

ADENOSINE-5′-DIPHOSPHATE causes reversible aggregation of blood platelets in many mammalian species both in vitro1,2 and in vivo3,4. An optimal concentration of calcium ions5 and the presence of one or more plasma factors6 are required for aggregation to take place. Studies by Born and Cross2 of the aggregation of human platelets in vitro have shown that there is a linear relationship between the rate of aggregation and the logarithm of the concentration of ADP. Aggregation in vitro, caused by ADP, is inhibited by adenosine, adenosine-5′-phosphate (AMP) and adenosine-5′-triphosphate (ATP) whether the platelets are human2,7, rabbit8 or pig5. It has been suggested by Clayton et al.9 that adenosine and AMP inhibit aggregation by occupying the ADP receptor.
