Poorly metastatic melanoma cell lines (human A375P or mouse B16F0) were injected intravenously into the tail veins of host mice and pulmonary metastases were isolated. Either these metastases were minced and grown in tissue culture (to be injected into additional host mice) or RNA was extracted to prepare the labelled cRNA used to hybridize to the oligonucleotide arrays. The procedure to select for highly metastatic tumour cells was repeated two (A375) or three (B16) times. A375SM cells were previously derived in a similar manner11.
