Upregulation of TLR2 protein expression in activated human HSCs in response to TNF-α or IL-1β. (a) Quantitation of TLR 2 and CD14 protein expression for different periods of time after TNF-α (10 ng/ml) or IL-1β (5 ng/ml) stimulation in activated HSCs was done by Western blotting. (be) Immunofluorescent staining was used to assess TLR2 protein expression in activated human HSCs. Cells were cultured in DMEM without serum. Cells were treated for 24 h as follows: (b, c) serum-free DMEM, (d) TNF-α (10 ng/ml), and (e) IL-1β (5 ng/ml). After 24 h of stimulation, (b) cells were fixed and stained with secondary antibody only, or (ce) cells were stained with rabbit anti-human TLR2 primary antibody and goat anti-rabbit rhodamin-conjugated secondary antibody.
