A, Effects of DEX and FK506 on COX-2 protein expression. Rheumatoid synovial fibroblasts were pretreated with FK506 for 2 hours. The cells were stimulated with IL-1β (5U/ml) in the presence of 10−9 M of DEX for 12 hours. The cells were lysed and lysates were analyzed by anti-COX-2 immunoblot. A representative example of three RA patients showing similar results. B, Results were calculated as a relative unit by densitometer. IL-1β control was assigned the value of 100%. Data were expressed as the percentage of IL-1β control.
