Flow cytometry comparisons of TLD-4A2, MHC class II, VLA-4, and ICAM-1 Ag expression on the EP T cell line. (The EP T cells were 94% TcR α, β+, and 95% CD4+ as assessed using flow cytometry.) The EP T cell line was stained under resting tissue culture conditions or after 3 days of ConA (5 μg/ml) activation in the presence of irradiated spleen accessory cells. Cells (1 × 106) were stained with a secondary isotype antibody control (20 Ab), TLD-4A2 mAb (7 μg/ml), OX-6 (MHC II) (50 μl of hybridoma supernate), anti-VLA-4 (10 μg/ml), and ICAM-1 (6 μg/ml) followed by goat anti-mouse IgG-FITC (1:650). Ten thousand events per stain were accumulated. Data demonstrate that the 4A2 molecule is expressed on more than 95% of EP T cells, lymph node cells, and cells from the thymus. The level of 4A2 expression is higher than that of MHC class II, VLA-4, and ICAM-1 on the same cells analyzed. Results presented here are representative of n = 2 flow cytometry experiments.
