Real-time PCR analysis of fibulin 2, P100, and IL-6 expression in peribiliary myofibroblasts and in myofibroblastic HSC. Total RNA (1 μg) from peribiliary myofibroblasts (closed symbols and bars) and from myofibroblastic HSC (open symbols and bars) at passages 1 to 4 (P1-P4) was reversed transcribed. Real-time PCR of target and 18S cDNA fragments was performed. A, Normalized emission-fluorescence (ΔRn) is plotted against cycle number. Threshold is shown. Ct value is determined as the cycle number at which the curve crosses the threshold. B to D, The data shown in A are expressed as the relative amounts of target transcripts in each sample, using myofibroblastic HSC at P1 as a control.
