The Gli-1 protein binds to DNA to cause upregulation of the expression of transforming growth factor-β (TGF-β). TGF-β then migrates through holes in the nuclear envelope into the extracellular space to cause proliferation of fibroblasts and upregulated expression of metalloproteinases by them that cause dissolution of basement membrane material.
