Sequence analysis of cDNA from SYT-SSX transcripts. RNA from synovial sarcoma specimens was used to generate RT-PCR amplicons using the SYT exon 10 sense primer in combination with an antisense SSX exon 5 primer annealing to a region downstream of the genotype specific SSX sequences. The amplicons were ligated into a plasmid vector and sequenced in both the forward and reverse directions. (a) Results from an SYT-SSX1 tumor; (b) results from an SYT-SSX2 tumor. The fusion junctions of the SYT and SSX exons are indicated. Nucleotides which define the SYT-SSX genotype are indicated above each sequence and are enclosed in boxes.
