Effect of increasing concentrations of zoledronic acid (ZOL) on MDA-MB-231 cell invasiveness through Matrigel. MDA-MB-231 cells were treated for 18 h by indicated concentrations of ZOL. After detachment and two washes in PBS, 2 × 105 cells were added in the upper Transwell chamber coated with Matrigel, as described in the Materials and Methods section. After 18 h incubation at 37°C, the cells in the upper part of the invasion chamber were gently detached and cells that had traversed the filter were counted by light microscopy after May–Grünewald–Giemsa coloration. In all, 10 fields (magnification × 200) were counted for each insert. Data are expressed as the percentage (as compared to control) of the mean±s.e.m. of five independent experiments.
