Zoledronic acid (ZOL) inhibits the chemotactic effect induced by SDF-1 on MDA-MB-231 cells – Partial reversion by geranylgeraniol (GGOH). Confluent MDA-MB-231 cells were treated for 18 h with 5 μg ml−1 exoenzyme or 10 μ M FTI-277 or 10 μ M GGTI-298 or 1 μ M ZOL in the presence or absence of GGOH and farnesol (FOH) and seeded into the upper chamber of Transwell coated with Matrigel, and a gradient of SDF-1 (100 ng ml−1) was established by placing the chemokine in the lower chamber. Then the invasion was measured as described in Figure 2. The inhibitory effect of ZOL was compared with that of C3 exoenzyme and of neutralising antibody against CXCR-4. Results are expressed as the percentage (as compared to control) of the mean±s.e.m. of five independent experiments.
