Etoposide-mediated effects on clusterin expression in PNT1a and PC-3 cells. Western blot detection of clusterin expression, cleaved caspase-3 and cleaved PARP in PNT1a (A) and PC-3 (E) cells. Equal protein loading was confirmed by pan-cytokeratin detection. Lanes 1: parental untransfected cells; lanes 2: parental cells treated overnight with 200 μ M etoposide; lanes 3: parental cells treated overnight with 400 μ M etoposide. PNT1a and PC-3 treated overnight with 400 μ M etoposide (phase images B and F) were tested for Annexin-V staining (respectively, C and G) and clusterin immunostaining (respectively, D and H).
