The role of histone deacetylation in silencing BNIP3 gene expression. (A) Effects of DNA methyltransferase and/or histone deacetylase inhibitor on the expression of BNIP3. SupT1 cells, which show BNIP3 methylation, were treated with 0.2 μ M 5-aza-dC, 300 nM TSA or both. (B) Histone acetylation examined by ChIP followed by PCR. Chromatin immunoprecipitation analysis was carried out using DNA precipitated with antiacetylated histone H3 antibody; the bars show the levels of histone acetylation determined by real-time-PCR normalised to the GAPDH signal.
