Small interfering RNA-B (siRNA-B) inhibition of PMA-induced angiogenesis on 3-D collagen gel. HUVE cells, seeded on 3-D collagen gels, were transfected with siRNA-B and siRNA-scrambled (C and D respectively; final concentration 200 pM) and treated for 48 h with PMA 40 nM in order to stimulate the early formation of capillary-like tubular structures. Results were compared with negative control (no PMA treatment, A) and PMA-stimulated positive control (B). The graph in (E) shows the number of capillary structures formed for each sample after treatments. All procedures are described under Material and Methods and results are expressed as mean±s.e.m. of three different experiments. #(P<0.05). Bar: 20 μm.
