The HBL cells were cultured in standard monolayers or in MCTS (30 000 cells) for 3 days. Supernatants were then harvested and their lactic acid content was measured as described in ‘Materials and Methods’. (B) Effects of lactic acid on antigen recognition by CTL. Cytotoxic T-lymphocyte from a MelanA/MART-127–35-specific HLA-A*0201 restricted clone were co-incubated with HBL melanoma cells cultured in 2D (□) or 3D (▪), as indicated, in the presence of lactic acid at the indicated concentrations.
