(AC) PKC-specific dye rim-1 indicates that a translocation of PKCs at diabetogenic conditions can be reversed by Go6976. (A) Depicts untreated MDA MB468 cells embedded in a 3D collagen matrix. As indicated by rim-1 staining, the PKC is localised in the cytosol, whereas Sytox green was used for staining of the nucleii. Rim-1 staining of MDA MB468 cells stimulated with 11 mM glucose and 100 ng ml−1 insulin shows a translocation of PKC towards the membrane and the pseudopodia – indicated by the arrows in (B), whereas the PKCα-specific inhibitor Go6976 (6 nM) was able to diminish the insulin-mediated translocation (C).
