  2. The T98G glioma cells were cultured as monolayer in DMEM with 10% FBS or as gliospheres in NBM with B27 and EGF+bFGF. After 5 days, the glioma cells were harvested using trypsin+EDTA, and gliospheres were dissociated using Accutase. Total RNA was extracted from glioma and BTSCs, and reverse transcribed into cDNA using random hexamer primers and TaqMan reverse transcription kit. The gene expression profile was determined using TaqMan Universal Master Mix with nuclear receptor primer sets and probes in an optical 384-well card using the 7900 Fast Sequence Detection Real-time PCR System (Applied Biosystems). The results were analysed using the Prism 7900 relative quantification (delta delta Ct) study software. The levels of nuclear receptor gene expression are normalised to 18 s, and the values are presented as fold change in BTSCs compared with glioma cells.
