AZA treatment of EBV-positive BL cells resulted in robust increase in PRDM1 mRNA. (a) EBV-positive and EBV-negative BL cell lines were treated with AZA and the levels of PRDM1α mRNA were measured by quantitative RT-PCR. The results were expressed relative to DMSO-treated BL cells (DMSO-treated =1), The mean values (±s.e.) from three independent experiments are shown. (b) Average methylation percentage of each of the 41 CpGs within the 5′ region of PRDM1 determined by bisulfite sequencing is shown for Mutu I. (c) Induction of EBV latency III-associated LMP1 and EBNA2, and decrease in BCL6 upon AZA treatment of Mutu I cells, determined by immunohistochemistry on cell block materials.
