POMC and cytokeratin (CK) release in a panel of 16 SCLC cell lines. All SCLC cell lines were seeded at 5 × 10⁴ ml⁻¹. Supernatants were taken at day 10 and POMC quantified by ELISA (developed in our lab) and CK by M65 ELISA (Peviva). Samples under the detection limit of the assay (15 pmol/l POMC and 11 U l⁻¹ CK) are displayed as 0. Results represent three independent cultures for each cell line and were assayed in duplicate and normalised to 10⁶ total cells. NCl-H526 cells have previously been shown to release CK, but not POMC. CK18 gene expression in SCLC cell lines. RNA was extracted from cell pellets grown in RTISS media for 10 days. NSCLC cells A549 were used as a positive control. GAPDH was used as a control for consistent mRNA levels between cell lines. CK18 band is located at 458 bp. All results are representative of three independent cultures of SCLC cell lines.
