APR-246 activates p53 target genes. (A) Changes in mRNA expression of p53 pathway and pro-apoptotic genes as measured by real-time quantitative RT–PCR, shown as fold induction relative to untreated cells in RM82 and STA-ET-7.2 cells after 24 h APR-246 treatments. (B) Induction of p21 (cdkn1A) and Puma (BBC3) mRNAs on treatment with 20 μM APR-246 in the p53 wild-type cell line TC252 but not in the p53-null cell line A673 as assessed by real-time PCR. (C) p53 knockdown by siRNA abrogates the induction of p21 mRNA by APR-246 in the STA-ET-7.2 cell line. Histograms represent the mean±s.e.m. of three independent experiments. A P value <0.05 is considered to be statistically significant.
