mBRAF increases MMP-1 and inflammatory cytokine levels and PLX4032 abrogates this increase. Bowes cells with empty vector (WT BRAF, clone 8) or BRAFV600E (mBRAF, clones 7 and 21) were incubated in serum-free medium (SFM) or in SFM containing 3 μ M PLX4032 (PLX) for 48 h. Real-time RT–PCR was used to measure the mRNA levels of (A) IL-1β, (B) IL-8, (C) IL-6, and (D) MMP-1. RT–PCR data were normalised to B2M, analysed by the 2ΔΔ(Ct) method (values are relative to clone 8 SFM) and are representative of three experiments. *P⩽0.05 compared to clone 8. #P⩽0.05 compared to same clone but with PLX4032 treatment.
