Forced expression of EGR3 induces IL6 and IL8 in P69 cells. (A) Early Growth Response 3 transfection in P69 cells. Cells were transfected with a tag-EGR3 expression plasmid or an empty plasmid as described in the Materials and Methods. Transfected cells were lysed and EGR3 protein expression was analysed by western blotting using antibodies against the V5 tag. (B) Subcellular localisation of EGR3 in transfected P69 cells. Untreated cells were lysed and fractionated using a commercial kit, followed by western blotting. (C) Total RNA was extracted from control or EGR3-expressing P69 cells and the mRNA expression of various EGR3-correlated genes was analysed by real-time qPCR. The housekeeping gene GAPDH was used as reference. Expression values were compared using the 2−ΔΔCT method.
