Conditioned media from juxtacrine culture accelerated cell migration and increased cell number of BMSCs. (a) Transwell migration assays of mouse BMSCs cultured for 8 hours in conditioned media from co-culture of macrophages and BMSCs, macrophage culture alone or BMSC culture alone. The migrated cells were counted after fixation and hematoxylin staining. Five fields of view (FV) were randomly picked up for each sample, and number of migrated cells per FV was counted. Quantitative results showed conditioned medium from co-culture of BMSCs and macrophages enhanced the migration of BMSCs. All experiments were performed in duplicate. Data are mean ± SEM (n = 2 in each group); *P < 0.05. (b) Image of BMSCs after fixation and hematoxylin staining in the Transwell assays. Images were taken at ×200 magnification. The remaining distance of scratch in BMSC cultures with conditioned media from co-culture of BMSCs and macrophages was significantly smaller than the other two groups.
