Tau is indispensable for WWOX regulation of neuronal differentiation. (a) SH-SY5Y cells were transfected with Tau siRNA, GSK3β siRNA and WWOX together or separately for 24 h. They were then treated with 10 μM RA for another 24 h. The photos were taken on day 1. (b) The percentage of differentiated cells with neurites longer than twice the length of the cell body was calculated. (c) Cells treated with Tau and GSK3β siRNA and WWOX plasmid were lysed in 2 × sample buffer, and a western blot analysis was carried out using antibodies against Tau, WWOX and GSK3β. (d) Our proposed model, showing that WWOX abolishes GSK3β kinase activity and therefore restores the ability of Tau to promote microtubule assembly
