Exogenous Wif1 suppressed Wnt-β-catenin signaling in the developing midline urorectum. β-gal-positive cells in TopgallacZ/+ embryos were localized at the distal urethral epithelium at E12.5 (a) and E13.5 (b). GT explant from the TopgallacZ/+ embryos at E12 were cultured for 24 h with increasing concentrations of Wif1 protein. The number of β-gal-positive cells (arrowheads) decreased in explants cultured with higher concentration of Wif1 proteins (c). GT from the E12 TopgallacZ/+ embryos cultured for 36 h with exogenous Wif1 (1.5 μg/ml) displayed cloaca membrane disintegration and un-septated cloaca (e) compared with the control (d).
