Loss of Bim plus Bmf rescues B-cell loss in TACI-Ig transgenic mice. (a) Splenocytes derived from mice of the indicated genotypes were stained with relevant fluorochrome-labeled monoclonal antibodies to distinguish T1 (IgM+ CD21), T2 (IgM+ CD21+), FO (CD23+ CD21+) and MZ (CD23 CD21+) B cells. Cell death in situ was monitored by Annexin V/7-AAD staining and flow cytometric analysis. (b) Different B-cell subsets were identified by flow cytometric analysis. T1 (IgM+CD21CD23), T2 (IgM+CD21+CD23), FO (CD21+CD23+), MZ (IgD+gMlowCD93CD1d+), PC (CD138+IgM−/low). Relative percentages and total splenic cellularities were used to calculate the absolute numbers of different B-cell subsets. Bars represent numbers 8–11±S.E.M. Significant differences are marked with *P<0.05; **P<0.005 and are compared with the genotypes±the TACI-Ig. # indicates significant differences P<0.05 between, wt, Bmf- and Bim-mutant mice expressing TACI-Ig
