Figure 3

Lm-infected p47phox−/− CD8+ memory lymphocytes express more pro-apoptotic Bim protein. Twenty-four hours (a) and 10 days (b) post reinfection freshly isolated splenocytes from WT and p47phox−/− mice were fixed and stained, and then analyzed for CD8, CD127, CD62L, and OVA257-264-tetramer expression. The data are the mean (±S.E.M.) percentage for six individual mice from two separate experiments with three of each genotype/experiment. **P<0.0001. Single-cell splenocyte cultures from WT and p47phox−/− mice infected with (c) 5 × 104 CFU (0.1 LD50) rLM-OVA, or (d) reinfected with 5 × 106 CFU (10 × LD50) for 24 h were fixed and stained, and then analyzed for CD8, CD127, CD62L, and intracellular Bim expression by flow-cytometry. The percentage of CD8+ lymphocytes expressing Bim is shown. The data are the mean (±S.E.M.) percentage for six individual mice from two separate experiments with three of each genotype/experiment **P<0.005
