Short- and long-term exposure to vorinostat induces autophagy in U937 cells. U937-B8 cells cultured in 2 μM vorinostat and their parental counterpart U937 (treated or not with 2 μ M vorinostat), each in the presence or absence of 25 μM CQ for 18 h, were fixed and imaged by TEM (a) or analyzed by immunofluorescence microscopy using an antibody specific for LC3 (right panel) (b). U937-B8 cells cultured in 2 μM vorinostat and parental U937 cells were treated or not with the indicated concentration of CQ for 18 h before total protein extracts were analyzed by western blotting using the antibodies indicated on the right (c and d). The atg5 antibody detects the higher molecular weight conjugate with atg12. U937 cells treated with 2 μ M vorinostat for the indicated times followed by western blotting of total protein extracts with the antibodies indicated on the right (e)
