Abnormal in situ ETC complex activities in Drp1 cells. (a) Complex I activity. In situ staining for NADPH (nicotinamide adenine dinucleotide phosphate reduced) ubiquinone oxidase was performed as previously described6 with gels similar to those shown in panel (a). A typical result is shown. (b) Quantification of the results shown in panel (a). The mean values of three independent in situ determinations are shown±1 S.E. Note the better resolution of the individual components of the supercomplexes (SC1 and SC2) than is seen in panel (a). (c) Complex V activity. In situ ATPase activity was determined as previously described6 using the same gels used to quantify Complex I activities. (d) Quantification of the results shown in panel (c).
