Metabolic abnormalities in Drp1 cells. (a) Quantification of OXPHOS. Vector and Drp1 cells were analyzed in parallel using a Metabolic Flux Analyzer as previously described.4 Note the higher basal OCR in vector cells as measured between 0 and 30 min (P<0.001). The addition of specific metabolic inhibitors at various times is indicated. (b) Quantification of glycolysis. ECAR was measured on the same samples used to quantify OCR in panel (a). Basal rates were significantly higher in Vector cells (P<0.001). (c) Severe depletion of ATP in Drp1 cells. ATP levels were determined as previously described4 and are indicated as the mean of quadruplicate samples±1S.E.
