Effect of TUG1 on cell growth in vitro. (a) The relative expression level of TUG1 in SPC-A1 cells, transfected with si-NC or si-TUG1 (si-TUG1#1, #2 and #3), was tested by qPCR. (b) At 24 h after transfection, MTT assay was performed to determine the proliferation of SPC-A1 and NCI-H1650 cells. (c) Representative results of colony formation of SPC-A1 and NCI-H1650 cells transfected with si-NC or si-TUG1. (d) At 48 h after transfection, cell cycle was analyzed by flow cytometry. The bar chart represents the percentage of cells in G1–G0, S, or G2–M phase, as indicated. (e) SPC-A1 transfected with si-NC/si-TUG1/p53+si-NC and transfected with p53 followed by transfection with si-RNA TUG1.
