Calpain mediates ESAT-6 induced neutrophil necrosis. (a) Pre-treatment of neutrophils with the calpain inhibitor, PD150606, inhibits the induction of necrosis by ESAT-6 (20 μg/ml). Necrosis was measured by flow-cytometry of propidium iodide uptake 1 h post-ESAT-6 stimulation. Data are normalised to vehicle plus DMSO. N=4 (in triplicate). *P<0.05.
