Influence and deletion of a conditional Bimfl allele in mice. (a) Lysates from splenocytes from wt, floxed Bim (Bimfl/fl) and BIM-deficient mice (Bimfl/fl/del) were immunoblotted for BIM. Probing for β-actin served as a loading control. N=2 mice per genotype were tested, six independent experiments were performed. (b) White blood cell counts and (c) spleen weights from wt, Bimfl/fl and Bimfl/fl/del mice were determined. N=5–6 mice for each genotype. Data represent mean±S.E.M. *P<0.05, **P<0.01, ***P<0.001 (paired t-test). (d) Thymocytes from mice of the indicated genotypes were isolated and single-cell suspensions stained with anti-CD4-PE (clone# YTA3.2.1) and anti-CD8-FITC (clone# 53.6.7.2). Dead cells were excluded from analysis by staining with 10 μg/ml propidium iodide (PI; Sigma-Aldrich)
