Thiostrepton induces apoptosis of SCs present in HT-29 and HCT-15 lines. (a) HT-29 and r29 cells were treated without (Ctrl) or with 5 and 10 μM thiostrepton for 48 h before their DNA contents were analyzed by flow cytometry. The percentage of the sub-G1 populations were indicated by the numbers shown on the left of each panel. (b) Both HT-29 and r29 cells after being treated with vehicle (Ctrl) and 5 or 10 μM thiostrepton for 48 h were collected for annexin V and 7-AAD staining. The apoptotic populations (numbers in the top right quadrant) were analyzed by flow cytometry. (c) Total lysates prepared from parental and Snail-overexpressing (Snail OE) HCT-15 cells as well as wild-type HT-29 and r29 cells after they were treated with or without the indicated concentrations of thiostrepton for 48 h were subjected to immunoblot analysis using an antibody against cleaved PARP as a probe. Tubulin signal served as a loading control
