miR-612 directly regulated AKT2 expression in CRC cells. (a) The wt, mutated, or deleted AKT2 3′-UTR was transfected into CRC cells with or without synthetic miR-612 mimic. (b) Luciferase activity was determined 36 h after transfection. Data were normalized to the luciferase activity transfected with miR-control (n=3). (c) Expression of AKT2 and p-AKT2 upon miR-612 overexpression or miR-612 inhibition was detected in two CRC cell lines by western blotting. (d) Relationship of relative miR-612 and AKT2 expression.
