From: Apoptotic cell clearance of Leishmania major-infected neutrophils by dendritic cells inhibits CD8+ T-cell priming in vitro by Mer tyrosine kinase-dependent signaling

Figure 7

Antibody blockade of Mer rescues the antigen presentation function of DCs. (a) C57BL/6 DCs were pretreated with isotype control or anti-Mer antibody for 1 h, then cultured or not with eGFPhiRFP− and eGFPhiRFP+ dermal neutrophils for 12–16 h. Sorted CD11c+, CD11c+GFP− and CD11c+GFP+ cells were cultured with CFDA-labeled OT-I CD8+ T cells for 3 days in the presence of OVA antigen. Representative histogram plots of CFDA fluorescence of CD8+CD3+ gated cells showing the proliferative response. Numbers represent the frequency of cells with reduced CFDA content. The data are representative of two independent experiments. (b) Gas6 mRNA expression in uninfected eGFPhiRFP− and L. major-infected eGFPhiRFP+ neutrophils purified from a total of 6 independent sorting procedures, each combining dermal cells pooled from 10 ears, and shown as relative expression levels in paired samples or as fold change in relative expression levels (mean±1 S.D.)
