ZnCl2 impairs NF-κB nuclear localization in HG condition. (a) RKO cells were grown in HG condition for 24 h with or without ZnCl2 (100 μM) before being assayed for immunocytochemistry with anti-NF-κB antibody. Nuclei were stained with Hoechst. (b) Western blotting and relative quantification of nuclear NF-κB/Lamin A ratio in RKO cells grown in HG condition for 24 h with or without ZnCl2 (100 μM) and ADR (2 μg/ml) treatment. Data represent mean±S.D. (c) Cells treated as in (a) were assayed for semi-quantitative RT-PCR of Twist-1 gene; 28S was used as a control for efficiency of RNA extraction and transcription.
