DR5 upregulation by CK was related to autophagy. HCT116 cells were pretreated with (b) or without (a) the autophagy inhibitor 3-MA, LY294002 (c) or transfected with Atg7 siRNA (e) and then exposed to CK at the doses indicated for 24 h. Whole-cell extracts were prepared and analyzed by western blotting using protein-specific antibodies. HCT116 cells were pretreated with 3-MA for 1 h (d) or transfected with Atg7 siRNA (f) and then treated with CK and/or TRAIL for 24 h. Cell death (as indicated by sub-G1 DNA content to the left of the G1 peak) was measured by FACS (d and f). Error bars in (d and f) represent the S.D.
