The increased chemoresistance of MSC-cocultured T-ALL cells is due to reduced mitochondrial ROS levels. (a) ROS generation was significantly decreased in primary T-ALL cells subjected to direct or indirect coculture with MSCs compared with mono-cultured. (b) Total superoxide production in primary T-ALL cells cultured with or without MSCs was measured using the Diogenes probe. (c) The Nox gene expression levels of primary T-ALL cells cultured with or without MSCs were examined by qRT-PCR. (d) MSCs coculture significantly decreased the generation of mitochondrial ROS in T-ALL cells. (e) Cells were treated with MitoTEMPO and the chemotherapeutic agents, and apoptosis was measured by Annexin V/PI staining and FACS.
