From: The role of autophagy in asparaginase-induced immune suppression of macrophages

Phagocytosis and proliferation are inhibited by asparaginase in Ana-1 and RAW264.7 cells. (a) Ana-1 and RAW264.7 cells were treated with 300 IU/ml IFN-γ and 200 ng/ml LPS, either alone or in combination with 0.1 IU/ml asparaginase for 24 h. The cells were incubated with zymosan particles for another 2 h, and analyzed by confocal fluorescent microscopy. (b) Ana-1 and RAW264.7 cells were labeled with CFSE, then incubated with 300 IU/ml IFN-γ and 200 ng/ml LPS, either alone or in combination with 0.1 IU/ml asparaginase for 24 h. The fluorescence intensity was analyzed by flow cytometry. Cells without treatment were used as control. (c) Ana-1 and RAW264.7 cells were treated with 300 IU/ml IFN-γ and 200 ng/ml LPS, in the presence or absence of 0.1 IU/ml asparaginase for 24 h, cell cycle distribution were analyzed by flow cytometry.
