DHA promotes activated HSC senescence in vitro. Primary HSCs were exposed for 6 h to 20 ng/ml PDGF-BB followed by vehicle, Etoposide or DHA treatment at indicated concentrations for 24 h. (a) Immunofluorescence using antibody against p53, p16, p21 and Hmga1. DAPI was used to stain the nucleus. Scale bars are 50 μm. (b) Senescence β-galactosidase staining was used to detect senescence. (c and d) Western blot and Real-time PCR analysis were used to determine the expression of Hmga1, p53, p21 and p16. Representative blots were from three independent experiments.
