Foxj3 and Zbtb18 are the targets of miR-219. (a) 3′-UTR analysis of Foxj3 and Zbtb18 containing putative regions that match the seed sequence of miR-219. (b) At 24 h after NIH/3T3 fibroblast cells were transfected with miR-219 mimics, luciferase reporter constructs containing WT or MUT-type UTRs were transfected as indicated. Cell lysates were harvested for DLR assays. (c–e) MiR-219 mimics or inhibitors were transfected to ESCs. After 48 h, cells were harvested for qRT-PCR (c) and western blot (d) to detect the relative levels of Foxj3 and Zbtb18. The relative intensities of the protein bands (e) were quantified with Image J software and calculated using the samples normalized to β-actin. All data are presented as mean±S.D. and derived from three independent experiments. Scale bars, 30 μm. **P<0.01
