Knockdown of HIF-1α using siRNA transfection abolished the chronic hypoxia upregulated Orai2 expression, while not affecting the upregulation of Orai1 in rat distal PASMCs. Six hours after transfected with non-targeting (siNT) or siRNA targeting HIF-1α (siHIF-1α), PASMCs were treated with prolonged hypoxia (4% O2, 60 h).
