FKBP52 affects the formation of complexes formed after addition of p23. (a) Incubation of an excess of FKBP52 in a solution containing Hsp902, Hsp702, Hop, GR (all at 1 μm), Hsp40 (0.3) μM and ATP (200 μM) leads to the formation of the predominant complex (Hsp90)2(Hop)1(Hsp70)2(GR)1(FKBP52)1. (b) Addition of p23 to this complex leads to formation of (Hsp90)2(GR)1(FKBP52)1 in an extended charge state series (highlighted in blue) indicative of two conformations, the higher m/z series is consistent with nucleotide binding. A very low intensity series of peaks is assigned to a population of (Hsp90)2(GR)1(FKBP52)1(p23)1 (red diamonds). A full assignment of this spectrum is available in Supplementary Figures S6 and S7.
