PBR1 knockdown impairs biogenesis of NADH dehydrogenase (NDH), photosystem I (PSI) and cytochrome b6f (Cytb6f) complexes. (a) Analysis of thylakoid membrane protein complexes by Blue-Native polyacrylamide gel electrophoresis (PAGE). Thylakoid membranes were isolated from wild-type (WT) and pbr1-1 mutant leaves and equal amounts of samples (3 μg chlorophyll) were separated. Red arrows indicate the reduction of corresponding complexes in pbr1-1 mutant. (b) Immunodetection of thylakoid complex proteins. The blots were probed with antibodies against the indicated proteins, respectively. (c) P700 redox kinetics determined by measuring absorbance of P700 at 820 nm induced by far-red light (FR, 720 nm). (d) NDH activities monitored by chlorophyll fluorescence. Leaves were exposed to actinic light (AL, 50 μmol of photons m−2 s−1) for 5 min.
