De novo design of macrocycles

Macrocyclic peptides, with sizes between that of small molecules and proteins, exhibit unique physicochemical and biochemical properties and have the potential to address protein targets inaccessible to traditional therapeutic modalities. However, efforts are largely limited to variants of natural products and there are currently no robust methods for the de novo design of protein-binding macrocycles.
