The integrated stress response (ISR) is a central regulator of cellular homeostasis that responds to diverse stressors. Dysregulation of the ISR contributes to the pathology of diseases, including viral infections, cancer and neurodegeneration. Although small-molecule activators of the ISR exist, many exhibit cytotoxicity or adverse effects.
