Resveratrol suppress Nox1-mediated ROS generation. (A) Cells were cultured in 96-well plates, pretreated with resveratrol (5 µM), NAC (5 mM) or apocynin (0.2 mM) for 1 h, suspended in PBS and stimulated with LPS containing lucigenin and NADPH. Chemiluminescence was measured in RLU every 10 min over a period of 150 min. (B, C) Macrophages were incubated for 1-6 h with 100 ng/ml of LPS in the presence or absence of resveratrol (5 µM). At the end of the incubation period, total mRNAs were isolated and Nox1 mRNAs were analyzed by RT-PCR (B) or real-time PCR (C). The mRNA levels of Nox1 were normalized to the levels of β-actin mRNA.
