Soluble factors from MSCs affected IL-10 secretion of splenocytes. (A) 5 × 105 BALB/c and 5 × 105 γ-irradiated C57BL/6 splenocytes were used as responder and stimulator cells, respectively, in MLR for 72 h. (A) ELISPOT assays were performed with or without culture supernatant from BALB/c MSCs. BALB/c fibroblast culture supernatant and MLR culture supernatant were used as controls. 100 µl of each supernatant was added per well. (B) The concentration of IL-10 in MLR culture supernatant, MSC culture supernatant or MLR with MSC CS was measured by ELISA. ***P < 0.0001. ('ND' means not detected) (C) Blocking of IL-10 and IL-10 receptor interaction was performed in the absence or presence of MSC culture supernatant, BALB/c fibroblast culture supernatant or MLR culture supernatant. A neutralizing IL-10 or IL-10 receptor Ab (5 µg/ml, respectively) was used in the indicated groups.
